What Selank is

Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a seven-amino-acid peptide derived from tuftsin, an endogenous human immunomodulatory peptide. The C-terminal Pro-Gly-Pro fragment was added to extend half-life — native tuftsin is broken down within minutes.

It was registered as a prescription anxiolytic in Russia in 2009 for generalized anxiety disorder and asthenia. Outside Russia it is investigational and available through compounding pharmacies for off-label use.

Patients describe the subjective effect as 'the volume of background anxiety turned down' rather than sedation — making it usable during work or driving.

How it works

Selank's anxiolytic effect appears to be mediated by modulation of GABAergic tone — not by direct binding to the benzodiazepine site, but by upregulating GABA-A receptor expression and shifting the balance of inhibitory and excitatory signaling.

It also raises hippocampal BDNF (brain-derived neurotrophic factor), the same growth factor implicated in long-term effects of SSRIs and exercise. This BDNF effect is the likely mechanism behind reported cognitive benefits.

Selank affects enkephalin metabolism (it inhibits enkephalin-degrading enzymes), which contributes to mood and stress regulation through endogenous opioid tone — a mechanism distinct from any other approved anxiolytic.

Zozulia AA et al. (Zhurnal Nevrologii i Psikhiatrii, 2008) reported clinical efficacy of Selank in generalized anxiety disorder in a Russian clinical study — the basis for its 2009 registration.

Most published RCTs are Russian-language and pre-date Western registration trial standards. Western RCT replication is limited. Honest framing: real clinical use base, modest peer-reviewed Western literature.
