What BDNF Mimetic is

BDNF mimetics are small molecules engineered to activate the TrkB receptor — the primary receptor for BDNF — without being BDNF itself. The most-studied is 7,8-dihydroxyflavone (7,8-DHF), a naturally occurring flavone with direct TrkB agonist activity. Related compounds include R7 and other flavonoid analogs.

Strictly speaking BDNF mimetics are small molecules, not peptides. They are included in the kindr longevity library because they target the same pathway as several neurotrophic peptides and are commonly considered alongside them in cognitive protocols.

They are investigational. They are not FDA-approved. U.S. clinical use is off-label, compounded by licensed 503A pharmacies under physician prescription.

How it works

BDNF signals through the TrkB receptor to activate PI3K-Akt (cell survival), MAPK-ERK (plasticity), and PLCγ (synaptic transmission) pathways.

7,8-DHF and related TrkB agonists bind a distinct site on TrkB and activate the same downstream cascade — bypassing the need for the large BDNF protein to reach the brain.

In preclinical models this supports hippocampal long-term potentiation, dendritic complexity, and resilience to stress and neurotoxic insults.
